Characterization of B cells in synovial fluid and tissue from patients with JIA

چکیده
Results SF B cells were found to be significantly enriched in CD27+ switch memory (sm) 1 cells and in the recently identified isotype class switch memory (CD19+CD27IgG+IgA+) B cells (sm2) compared to paired and healthy PB (P < 0.0001). CCR5, CCR8, and CCR9 expression was significantly higher on SF sm1 and sm2 B cells than on correspondent paired PB B cells (P < 0.001). Naïve (IgD+, CD27-) B cells were significantly reduced in SF compared to paired and control PB (P < 0.0001). Similarly, transitional B cells (CD19+CD24highCD38highIgMhighIgDhigh) were significantly less numerous in SF than in paired PB from JIA patients (P < 0.0001).

Background: Heat-shock proteins are part of a strictly controlled biological system that allows organisms to respond to environmental stresses. Different proinflammatory cytokines are present in the synovial tissue of rheumatoid arthritis patients. Such tissues respond to stress and induce heat-shock proteins. In addition, synovial cells are exposed to mechanical stress caused by joint motion. ...
متن کاملPhenotypic and functional characterization of switch memory B cells from patients with oligoarticular juvenile idiopathic arthritis
INTRODUCTION In chronic inflammatory disorders, B cells can contribute to tissue damage by autoantibody production and antigen presentation to T cells. Here, we have characterized synovial fluid and tissue B-cell subsets in patients with oligoarticular juvenile idiopathic arthritis (JIA), an issue not addressed before in detail. METHODS B cells from synovial fluid (SF) and peripheral blood (P...

Background: Triggering receptors expressed on myeloid cells i.e soluble triggering expressed on myeloid cells-1 (STREM-1) is inducible on monocyte/macrophages and neutrophils and accelerates tissue destruction by propagating inflammatory responses in disease related to bacterial infections. The aim of this study was to investigate the role of STREM-1 in the synovial fluid to identify septic...

Total IgM, IgA, and IgM-rheumatoid factor (RF) and IgA-RF were quantitated in the serum and synovial fluid of 35 seropositive and 8 seronegative rheumatoid arthritis (RA) patients, using a sensitive enzyme-linked immunosorbent assay (ELISA). Mononuclear cells isolated from peripheral blood and synovial fluid of three seropositive patients were also stimulated in vitro with phorbol myristat...
متن کاملOsteoblast differentiation from synovial fluid cells in juvenile idiopathic arthritis (JIA)
Purpose Juvenile idiopathic arthritis (JIA) is one of the leading causes of disability in children characterized by destruction of articular cartilage and underlying bone, as well as synovial hyperplasia. Since synovia contains osteoblast progenitors, hyperplastic changes may contribute to joint destruction by inhibition of osteoblast differentiation. Osteoblasts are also immunoregulatory cells...
